Plasminogen activator in bile during extracorporeal perfusion of pig liver.

چکیده
Plasminogen activator activity was studied in bile and perfusate during extracorporeal pig liver perfusion. Plasminogen activator activity in bile was up to 58 times greater than in perfusate. A high level of plasminogen activator activity in bile was also found after the addition of urokinase to the perfusate. These results suggest that bile is the main route of active excretion of plasminogen activator and urokinase from the liver in experimental liver perfusion.

Normothermic extracorporeal perfusion of isolated porcine liver after warm ischaemia: a preliminary report.
Liver transplantation is a major life-saving procedure, and donation after cardiac death (DCD) has increased the pool of potential liver donors. However, DCD livers are at increased risk of primary graft dysfunction and biliary tract ischaemia. Normothermic extracorporeal liver perfusion (NELP) may increase the ability to protect, evaluate and, in future, transplant DCD livers. We conducted pro...

Porcine lactic dehydrogenase (L.D.H.) has been found in the serum of four patients in hepatic coma treated by extracorporeal porcine liver perfusion. A comparison between the porcine L.D.H. isoenzyme pattern in the patients' serum and that in porcine serum and liver extract indicates that the porcine L.D.H. is derived from the pig liver. This finding reflects damage to the pig liver cells susta...
متن کاملCritical role of plasminogen activator inhibitor-1 in cholestatic liver injury and fibrosis.
Plasminogen activator inhibitor-1 (PAI-1) is an acute phase protein known to correlate with hepatic fibrosis. However, whether or not PAI-1 plays a causal role in this disease process had not been directly tested. Therefore, wild-type or PAI-1 knockout (PAI-1(-/-)) mice underwent bile duct ligation. Mice were sacrificed either 3 or 14 days after surgery for assessment of early (i.e., inflammati...
متن کاملBlocking porcine sialoadhesin improves extracorporeal porcine liver xenoperfusion with human blood.
BACKGROUND Patients in fulminant hepatic failure currently do not have a temporary means of support while awaiting liver transplantation. A potential therapeutic approach for such patients is the use of extracorporeal perfusion with porcine livers as a form of "liver dialysis". During a 72-h extracorporeal perfusion of porcine livers with human blood, porcine Kupffer cells bind to and phagocyto...
متن کاملXenogeneic, extracorporeal liver perfusion in primates improves the ratio of branched-chain amino acids to aromatic amino acids (Fischer's ratio).
In fulminant hepatic failure (FHF), the development of hepatic encephalopathy is associated with grossly abnormal concentrations of plasma amino acids (PAA). Normalization of the ratio of branched-chain amino acids to aromatic amino acids (Fischer's ratio) correlates with clinical improvement. This study evaluated changes in PAA metabolism during 4 h of isolated, normothermic extracorporeal liv...
